Oral Hexarelin is a synthetic growth hormone–releasing peptide (GHRP) belonging to the hexapeptide family, known for its potent stimulation of endogenous growth hormone (GH) release. Like injectable Hexarelin, it acts by binding to the growth hormone secretagogue receptor (GHS-R1a) in the hypothalamus and pituitary gland, triggering pulsatile GH secretion and downstream increases in insulin-like growth factor-1 (IGF-1). These effects are associated with roles in metabolism, tissue repair, and body composition regulation.
However, oral administration of peptide-based compounds such as Hexarelin presents significant challenges. Peptides are generally susceptible to degradation in the gastrointestinal tract due to enzymatic digestion and poor membrane permeability, resulting in low oral bioavailability. Studies on GHRPs indicate that non-injectable delivery often leads to reduced systemic absorption compared to parenteral routes, limiting effectiveness unless specialized delivery systems are used.
Despite these limitations, research into oral or alternative delivery systems for growth hormone secretagogues continues, including formulations designed to enhance stability and absorption. Additionally, Hexarelin has demonstrated biological activity beyond GH release, including potential cardioprotective effects mediated through CD36 receptors in cardiac tissue.
Overall, while oral Hexarelin is conceptually appealing for ease of use, current evidence suggests that injectable administration remains the more reliable method for achieving consistent physiological effects.
