5-Amino-1MQ (5-amino-1-methylquinolinium) is an investigational small molecule that functions as a nicotinamide N-methyltransferase (NNMT) inhibitor, a key enzyme involved in cellular energy and NAD⁺ metabolism. By inhibiting NNMT, 5-Amino-1MQ reduces the conversion of nicotinamide into 1-methylnicotinamide, which may help preserve intracellular NAD⁺ levels and support mitochondrial function and metabolic regulation.
Preclinical research suggests that NNMT inhibition may influence adipocyte biology and energy balance. In animal and in vitro studies, NNMT inhibition has been associated with reduced fat accumulation, improved metabolic profiles, and changes in cellular energy utilization. For example, studies in diet-induced obese mice demonstrated reduced body weight and adipose tissue with NNMT inhibition, supporting its potential role in metabolic modulation (Kraus et al., 2018, PMC5826726). Additional research indicates that NNMT activity is linked to obesity and metabolic dysfunction, making it a potential therapeutic target (Ulanovskaya et al., 2013; Nature Scientific Reports, 2021).
Despite these findings, no human clinical trials have established safety, dosing, or efficacy for 5-Amino-1MQ. It remains an experimental compound used primarily in research settings. Therefore, its use in humans should be approached cautiously and under qualified medical supervision.
